Abstract The cell of origin Group 3 medulloblastoma, the most malignant medulloblastoma subgroup, is currently unknown and remains controversial. Transcriptional profiling has revealed that medulloblastomas are characterized by elevated expression a photoreceptor program, which not been described in normal cerebellar development but well developing retina pineal gland. We used lineage tracing single-cell sequencing to compare brain with tumor our previously generated MYC-driven transgenic mouse model (GMYC), where mice spontaneously develop after 4-6 months age. found cells emerged from progenitor MYC overexpression drove transformation immature pathway activity. Our data suggest originates expressing implications for future research novel treatments targeting this devastating childhood malignancy.
